Is it bad to be an anime fan or an otaku when it comes to being in a relationship?

Anime is most certainly not the problem. I know plenty of guys and girls who love anime and other nerd things and have tons of success in relationships, sex and all that stuff. Most of those friends would count me among that group but I have enough self awareness to realise I'm a bit of a dolt. As long as there's actual love, acceptance and even tolerance for flaws and quirks then that's okay. It's just that most people in nerd culture have, like what John Felix said, a crippling social dysfunction. But most people don't realise it's REALLY not that hard to talk to people but as long as you're trying your best to get what you want then I don't see an issue with being on the shy side. Just don't expect others to do the work for you.

^comparing the ratio of [successful to unsuccessful] anime couples to the ratio of [successful to unsuccessful] normie couples would be a better estimate. i am not against what you said, i also think being an otaku is not bad for a relationship. But what you said about 'finding successful otaku couples everywhere' does not prove anything, it can just be cherry picking.
